To investigate cell-specific transcriptomes, we clustered the dynamic patterns of mRNAs. A large proportion of genes had significant expression variations among monocytes, imDCs and mDCs. Genes with the same expression levels in either two of the three cell types rarely appeared, which implied heterogeneity of these cells. During DC differentiation and maturation, there were higher proportions of transcriptional up-regulated genes which were possible DC-specific functional molecules. The expression variation trends of known functional genes of DCs, such as human leukocyte antigen (HLA) molecules, co-stimulators and pro-inflammatory cytokines, are the same as previously reported, which implied high quality of our transcriptome data. In a word, these results indicate that DC-specific functional states are achieved through the dynamic transcriptional patterns to establish DC-specific functional status.
