Rare DICER1 and Absent FOXL2 Mutations Characterize Ovarian Juvenile Granulosa Cell Tumors
FOXL2 somatic mutation occurs in a high percentage of ovarian adult granulosa cell tumors and DICER1 mutations in a high proportion of Sertoli-Leydig cell tumors. These mutations have only been studied in a limited number of juvenile granulosa cell tumors (JGCTs), and their occurrence and frequency in these neoplasms is controversial. We aimed to determine the frequency of FOXL2 and DICER1 mutations in a large cohort of 50 JGCTs, and to evaluate the prognostic impact of these mutations. A FOXL2 hotspot...

Evolutionary history of dimethylsulfoniopropionate (DMSP) demethylation enzyme DmdA in marine bacteria
Dimethylsulfoniopropionate (DMSP), an osmolyte produced by oceanic phytoplankton and bacteria, is primarily degraded by bacteria belonging to the Roseobacter lineage and other marine Alphaproteobacteria via DMSP-dependent demethylase A protein (DmdA). To date, the evolutionary history of DmdA gene family is unclear. Some studies indicate a common ancestry between DmdA and GcvT gene families and a co-evolution between Roseobacter and the DMSP-producing-phytoplankton around 250 million years ago (Mya)....

A walk in the park? Characterizing gait‐related artifacts in mobile EEG recordings
Abstract Brain activity during natural walking outdoors can be captured using mobile electroencephalography (EEG). However, EEG recorded during gait is confounded with artifacts from various sources, possibly obstructing the interpretation of brain activity patterns. Currently, there is no consensus on how the amount of artifact present in these recordings should be quantified, nor is there a systematic description of gait artifact properties. In the current study, we expand several features into...

Basophil Activation Test : A diagnostic, predictive and monitoring assay for Allergen Immunotherapy
Abstract For decades, the approach to clinical care has involved the assessment of a fairly small set of patients' signs and symptoms, sampled over a short period of time with limited attention given to individual variations in aetiology and pathophysiology. Subsequently, a therapy is assigned predominantly with a "one shoe fits all" approach. Precision medicine approaches are now starting to change ways to test, treat and develop new therapies for allergic and immunological disorders. (Figure 1).

The heterogeneity of even “well-defined” type 2 (T2) asthma is reflected by the absence of uniform response to T2 blockade despite recruitment of subjects with a strong T2 signal. Fractional exhaled nitric oxide (FeNO) is a biomarker elevated in uncontrolled T2-asthma secondary to IL-13 induced nitric oxide synthase expression in the airway epithelium. While FeNO is reduced by anti-IgE and anti-IL-4/13 therapies(1-5), it did not change in studies with mepolizumab, suggesting the absence of modulation...

Immunologic and tumor responses of pegilodecakin with 5-FU/LV and oxaliplatin (FOLFOX) in pancreatic ductal adenocarcinoma (PDAC)
Summary Background Treatment options for pancreatic ductal adenocarcinoma (PDAC) are limited and checkpoint blockade inhibitors have been disappointing in this disease. Pegilodecakin has demonstrated single agent anti-tumor activity in immune-sensitive tumors. Phase 1 and preclinical data indicate synergy of pegilodecakin with 5-FU and platins. We assessed the safety and activity of pegilodecakin+FOLFOX in patients with PDAC. Methods IVY (NCT02009449) was an open-label phase 1b...

The Shifting Demographics of Birth Related Brachial Plexus Injury: The Impact of Socioeconomic Status and Ethnic group
Many of the risk factors for Birth Related Brachial Plexus Injury (BRBPI) such as maternal gestational diabetes and macrosomia, are known to vary between demographic groups. Socioeconomic differences are known to influence access to healthcare, including elective Caesarean section rates and access to consultant Obstetricians, which could impact rates of BRBPI. This study aims to explore whether BRBPI is affected by demographic factors.

Characterization of a Novel Entity of G3 (High-grade Well-differentiated) Colorectal Neuroendocrine Tumors (NET) in the SEER Database
Objectives: Small studies suggest that a new entity of high-grade (HG) (G3, by Ki-67 or mitotic index) well-differentiated (histologically) gastrointestinal neuroendocrine tumors (NETs) exists, but prognosis and characteristics are unknown. We further characterized demographics and prognosis of patients with colorectal G3 NETs. Materials and Methods: We used the Surveillance Epidemiology and End Results (SEER) database to study colorectal NETs diagnosed from 2000 to 2015. We evaluated...

Post-treatment neutrophil-to-lymphocyte ratio (NLR) predicts response to anti-PD-1/PD-L1 antibody in SCLC patients at early phase
Abstract Neutrophil-to-lymphocyte ratio (NLR), platelet-to-lymphocyte ratio (PLR), systemic immune-inflammation index (SII) have been identified as predictors of treatment response in a variety of cancers. We conducted a retrospective analysis to investigate the usefulness of NLR, PLR and SII at baseline and at 6 weeks post-treatment as predictors of response to anti-PD-1/PD-L1 antibody treatment in small cell lung cancer (SCLC). Data of 41 SCLC patients receiving immunotherapy...
