A FACS-based Protocol to Isolate RNA from the Secondary Cells of Drosophila Male Accessory Glands
Here, we present a protocol to dissociate and sort a specific cell population from the Drosophila male accessory glands (secondary cells) for RNA sequencing and RT-qPCR. Cell isolation is accomplished through FACS purification of GFP-expressing secondary cells after a multistep-dissociation process requiring dissection, proteases digestion and mechanical dispersion.

Use of Micro X-ray Computed Tomography with Phosphotungstic Acid Preparation to Visualize Human Fibromuscular Tissue
Micro X-ray computed tomography is effective in obtaining three-dimensional information from undamaged human specimens but has limited success in observing soft tissues. The use of phosphotungstic acid contrast agent can resolve this issue. We implemented this contrast agent to examine human delicate fibromuscular tissues (the orbicularis retaining ligament).

A Photonic System for Generating Unconditional Polarization-Entangled Photons Based on Multiple Quantum Interference
We describe an optical system for the generation of unconditional polarization-entangled photons based on multiple quantum interference effects with a detection scheme to estimate the experimental fidelity of generated entangled photons.

Examining Online Syntactic Processing of Spoken Complex Sentences in Chinese Using Dual-Modal Interference Tasks
Here, we present a protocol employing dual-modal interference tasks to examine online processing of spoken-Chinese relative clause sentences. Two exemplar experiments involving auditory processing with intra- and extrasentential interferences are described. The paradigm provides a methodology for addressing the nature of working memory and its effects on sentence processing.

Flow Cytometric Analysis of Mitochondrial Reactive Oxygen Species in Murine Hematopoietic Stem and Progenitor Cells and MLL-AF9 Driven Leukemia
We describe a method for using multiparameter flow cytometry to detect mitochondrial reactive oxygen species (ROS) in murine healthy hematopoietic stem and progenitor cells (HSPCs) and leukemia cells from a mouse model of acute myeloid leukemia (AML) driven by MLL-AF9.

Single-Molecule Tracking Microscopy - A Tool for Determining the Diffusive States of Cytosolic Molecules
3D single-molecule localization microscopy is utilized to probe the spatial positions and motion trajectories of fluorescently labeled proteins in living bacterial cells. The experimental and data analysis protocol described herein determines the prevalent diffusive behaviors of cytosolic proteins based on pooled single-molecule trajectories.

Inhaled GM-CSF for Pulmonary Alveolar Proteinosis
Pulmonary alveolar proteinosis, an uncommon lung disease characterized by the accumulation of pulmonary surfactant within pulmonary alveoli, causes progressive respiratory insufficiency. Approximately 90% of cases of pulmonary alveolar proteinosis are autoimmune; autoimmune pulmonary alveolar…

Five-Year Outcomes of a Randomized Trial of Treatments for Varicose Veins
Minimally invasive treatment options such as endovenous laser ablation therapy and ultrasound-guided foam sclerotherapy are now recommended alternatives to surgery for the treatment of varicose veins. Medium-term follow-up studies suggest that laser ablation, foam sclerotherapy, and surgery result…

Trial of SAGE-217 in Patients with Major Depressive Disorder
Antidepressants that primarily enhance monoaminergic neurotransmission involving serotonin or norepinephrine are used in the treatment of major depressive disorder, and their clinical effects are generally evident in 4 to 8 weeks. One hypothesis for the mechanism of depression implicates deficits…

Safety and Efficacy of Mitapivat in Pyruvate Kinase Deficiency
Pyruvate kinase deficiency is an autosomal recessive enzymopathy in red cells that is caused by mutations in PKLR. These genetic alterations lead to a deficit of pyruvate kinase activity in red cells and to hemolytic anemia of variable severity. In addition to anemia, pyruvate kinase deficiency is…

Baseline of upper teeth: (a) Control organ for spatial navigation? (b) Weak point for misaligned posture and pain?
Publication date: Available online 5 September 2019Source: Medical HypothesesAuthor(s): A. Kapp, M. Paethke, M. BaquéAbstractOur observations question both the current doctrine of spatial orientation as processed by vestibular, visual and proprioceptive impressions as well as the horizontal alignment of the eye axis. Indeed our observations suggest spatial orientation as a physically based, largely mechanically transmitted interaction between individual and environment. It is controlled by an interface...

Relating the Diers formetric measurements with the subjective severity of acute and chronic low back pain
Publication date: Available online 4 September 2019Source: Medical HypothesesAuthor(s): Inge Roman, Michiel Luyten, Hendrik Croonenborghs, Grégoire Lason, Luc Peeters, Geert Byttebier, Frank ComhaireAbstractBackgroundcontext: Nonspecific low back pain (LBP) is a common disorder with a high economic, social and psychological burden. Many systems have been developed for evaluating the severity of LBP, though these are mainly based on scoring questionnaires for the functional status of the patients....
