Prolyl-hydroxylase inhibitors for the treatment of anemia in chronic kidney disease
Purpose of review Prolyl-hydroxylase inhibitors are a novel class of orally administered drugs that are under development for the treatment of anemia in patients with chronic kidney disease. This review discusses the biology of these drugs and their target – hypoxia-inducible factor and potential advantages and disadvantages of these therapies. Finally, we will discuss current trials in patients with both chronic kidney disease and end-stage renal disease. Recent findings Recent smaller studies...

An RNAi Screen for Genes Required for Growth of Drosophila Wing Tissue
Cell division and tissue growth must be coordinated with development. Defects in these processes are the basis for a number of diseases, including developmental malformations and cancer. We have conducted an unbiased RNAi screen for genes that are required for growth in the Drosophila wing, using GAL4-inducible short hairpin RNA (shRNA) fly strains made by the Drosophila RNAi Screening Center. shRNA expression down the center of the larval wing disc using dpp-GAL4, and the central region of the adult...

Alternative Lengthening of Telomeres in the Budding Yeast Naumovozyma castellii
The enzyme telomerase ensures the integrity of linear chromosomes by maintaining telomere length. As a hallmark of cancer, cell immortalization and unlimited proliferation is gained by reactivation of telomerase. However, a significant fraction of cancer cells instead uses alternative telomere lengthening mechanisms to ensure telomere function, collectively known as Alternative Lengthening of Telomeres (ALT). Although the budding yeast Naumovozyma castellii (Saccharomyces castellii) has a proficient...

Exon 3 of the NUMB Gene Emerged in the Chordate Lineage Coopting the NUMB Protein to the Regulation of MDM2
MDM2 regulates a variety of cellular processes through its dual protein:protein interaction and ubiquitin ligase activities. One major function of MDM2 is to bind and ubiquitinate P53, thereby regulating its proteasomal degradation. This function is in turn controlled by the cell fate determinant NUMB, which binds to and inhibits MDM2 via a short stretch of 11 amino acids, contained in its phosphotyrosine-binding (PTB) domain, encoded by exon 3 of the NUMB gene. The NUMB-MDM2-P53 circuitry is relevant...

Laparoscopic versus open subtotal gastrectomy for adenocarcinoma of the stomach in a Western population: peri-operative and 5-year oncological outcomes
ConclusionThis study suggests that LSG is comparable to OSG in Western patients with respect to oncological quality and peri-operative morbidity. Two-year overall survival is predicted by the number of positive lymph nodes and not the operative access employed for resection. Performance of endobronchial-ultrasound guided miniforceps biopsy of targeted mediastinal and hilar lesions
Biopsy instruments are being developed to enhance the diagnostic yield of endobronchial ultrasound (EBUS) sampling. Convex linear probe EBUS is a modality that has been refined and become the primary approach for tissue acquisition, particularly for target lesions in the mediastinum and pulmonary hila. Using this technology, EBUS guided transbronchial needle aspiration (TBNA) has been shown to have high sensitivity and specificity in diagnosing small cell and non-small cell lung carcinomatosis, whether...

Loss of thymidine kinase 1 inhibits lung cancer growth and metastatic attributes by reducing GDF15 expression
by Parmanand Malvi, Radoslav Janostiak, Arvindhan Nagarajan, Guoping Cai, Narendra Wajapeyee Metabolic alterations that are critical for cancer cell growth and metastasis are one of the key hallmarks of cancer. Here, we show that thymidine kinase 1 (TK1) is significantly overexpressed in tumor samples from lung adenocarcinoma (LUAD) patients relative to normal controls, and this TK1 overe xpression is associated with significantly reduced overall survival and cancer recurrence. Genetic knockdown...

Clues from DNA could help predict growth of prostate cancer
FINDINGSResearchers from the UCLA Jonsson Comprehensive Cancer Center and other institutions in the U.S., Canada, the United Kingdom and Singapore, have identified 1,178 biomarkers in men ’s genomes — the complete set of genetic material inherited from one’s parents — that predict how an individual person’s prostate cancer will grow. The finding suggests that predicting how a person ’s cancer will evolve may lie in their inherited DNA.BACKGROUNDThe researchers focused in part on a biological process...

Keytruda for Mesothelioma Flops in Phase III Clinical Trial
Results from a phase III clinical trial comparing Keytruda (pembrolizumab) to standard chemotherapy shows the immunotherapy drug still has a long way to go as a viable treatment option for malignant pleural mesothelioma. Keytruda did not improve progression-free survival for mesothelioma patients who progressed after first-line chemotherapy. The disappointing results from the PROMISE-meso study were presented at the European Society of Medical Oncology (ESMO) annual meeting last week. It was...

On the accuracy of bulk synthetic CT for MR-guided online adaptive radiotherapy
Abstract Purpose MR-guided radiotherapy (MRgRT) relies on the daily assignment of a relative electron density (RED) map to allow the fraction specific dose calculation. One approach to assign the RED map consists of segmenting the daily magnetic resonance image into five different density levels and assigning a RED bulk value to each level to generate a synthetic CT (sCT). The aim of this study is to evaluate the dose calculation accuracy...

Impact of sarcopenia on postoperative surgical site infections in patients undergoing flap reconstruction for oral cancer
Sarcopenia is characterized by progressive and generalized loss of skeletal muscle mass and strength. The aim of the study was to investigate the impact of skeletal muscle mass on surgical site infection (SSI) in flap reconstruction for defects after oral cancer resection. The subjects were a non-randomized, retrospective cohort of 106 patients who underwent this procedure after preoperative abdominal-lumbar computed tomography (CT). Cross-sectional areas (cm2) of skeletal muscles in the L3 region...

Drug causes of intracerebral haemorrhage
Summary Many drugs have the potential to cause intracerebral haemorrhage. The mechanisms that underlie this association include elevation of blood pressure and increasing bleeding tendency. The consequences of intracerebral haemorrhage can be devastating for the individual, so careful risk assessment prior to drug initiation and close monitoring during treatment should be enacted when using medicines with an established association with haemorrhagic stroke.

Chiral checkpoints during protein biosynthesis [Cell Biology]
Protein chains contain only L-amino acids, with the exception of the achiral glycine, making the chains homochiral. This homochirality is a prerequisite for proper protein folding and, hence, normal cellular function. The importance of D-amino acids as a component of the bacterial cell wall and their roles in neurotransmission in higher eukaryotes is well established. However, the wider presence and the corresponding physiological roles of these specific amino acid stereoisomers have been appreciated...
