Gastrointestinal tract dysbiosis enhances distal tumor progression through suppression of leukocyte trafficking
The overall use of antibiotics has increased significantly in recent years. Besides fighting infections, antibiotics also alter the gut microbiota. Commensal bacteria in the gastrointestinal tract are crucial to maintain immune homeostasis, and microbial imbalance or dysbiosis affects disease susceptibility and progression. We hypothesized that antibiotic-induced dysbiosis of the gut microbiota would suppress cytokine profiles in the host, thereby leading to changes in the tumor microenvironment....

Inactivating NF1 mutations are enriched in advanced breast cancer and contribute to endocrine therapy resistance
Purpose:Advanced breast cancer (ABC) has not been subjected to the same degree of molecular scrutiny as early primary cancer. Breast cancer evolves with time and under the selective pressure of treatment, with the potential to acquire mutations with resistance to treatment and disease progression. To identify potentially targetable mutations in advanced breast cancer, we performed prospective molecular characterisation of a cohort of patients with ABC. Experimental Design:Biopsies from patients with...

A Molecular Study of Synovial Chondromatosis
ABSTRACT Synovial chondromatosis (SC) is a rare benign cartilaginous neoplasm in which recurrent FN1 and ACVR2A gene rearrangements have been recently reported. Triggered by a case of malignant transformation in SC (synovial chondrosarcoma) showing a novel KMT2A‐BCOR gene fusion by targeted RNA sequencing, we sought to evaluate the molecular abnormalities in a cohort of 27 SC cases using a combined methodology of FISH and/or targeted RNA sequencing. Results showed that FN1 and /or ACVR2A gene rearrangements...

The extended spectrum of RAS‐MAPK pathway mutations in colorectal cancer
Abstract Current clinical guidelines recommend mutation analysis for select codons in KRAS and NRAS exons 2, 3 and 4 and BRAF V600E to guide therapy selection and prognostic stratification in advanced colorectal cancer. This study evaluates the impact of extended molecular testing on the detection of RAS‐MAPK pathway mutations. Panel next‐generation sequencing results of colorectal cancer specimens from 5795 individuals from the AACR Project GENIE were included. Mutations in RAS‐MAPK pathway genes...
